This is the third video on the series: PHYLOGENETICS. This video is aimed at the analysis of Chromatograms obtained from Sanger Sequencing. In this video you will be given perspective on different analysis software to use, assessing the quality of Chromatogram sequences, editing Chromatograms and exporting the sequences in FASTA format for downstream analysis.

Should i have to do BLAST individually for all my Sanger sequencing data? If yes.. Then why should i do that? If i collect samples from known organiams.

Good question. The simple answer is about sequence quality and reliability of results. Keeping the ends of these sequences could mean that you are including inaccurate reads. This could lead to making the incorrect matches with your sequences. With Sanger sequencing the ends of the sequences are usually unreliable due to the nature of the sequencing technology and should be cut away. Remember to use the quality bars to assess how much should be trimmed.

Hi there! Yes, in this example you would perform two or three PCRs on a sample and sequence each of those samples individually. This should increase your degree of reliability by supporting accurate results and highlighting inaccurate results.
